什么是CRISPR?
CRISPR / Cas9是一种RNA导向的靶向基因组编辑工具,可让研究人员针对细胞系或动物进行基因敲除、SNP、插入和缺失。 CRISPR / Cas9基因组编辑系统需要两个组件,Cas9,核酸内切酶和向导RNA(sgRNA);sgRNA特异性地将Cas9引导至基因组序列中的位置。 在3'末端存在与原间隔物相邻的基序(PAM-序列NGG),Cas9将解开DNA双链体并在被引导RNA识别靶序列后切割两条链。
