Background | Mouse MIP-3α, otherwise known as CCL20, LARC, and Exodus, is an 8 kDa protein belonging to the β-, or CC chemokine sub-family. It is synthesized as a 97 amino acid (aa) precursor that contains a 27 aa signal sequence plus a 70 aa mature segment. Mature mouse MIP-3α contains no potential N-linked glycosylation sites. Although human MIP-3α is suggested to contain an O-linked ribose, rodent MIP-3α does not contain any proline rich motifs that might indicate glycosylation. MIP-3α is known to exist in multiple isoforms. An alternate splice form lacking the N-terminal alanine exists in both mouse and human. In addition, human MIP-3α apparently undergoes proteolytic processing at the C-terminus to generate peptides that are shorter by one, three, and six amino acid (aa) residues. All isoforms are equally bioactive. It is possible that mouse MIP-3α may be processed similarly. The mature region of mouse MIP-3α shares 73% and 70% aa sequence identity with mature rat and human MIP-3α, respectively. Among mouse β-chemokines, MIP-3α is distantly related to MIP-3β sharing 34% aa sequence identity. Cells known to express MIP-3α include monocytes, macrophages, microglia and astrocytes, mast cells, neutrophils, keratinocytes, intestinal epithelium, osteoblasts, fibroblasts, and endothelial cells. |
