Background | One of the first cytokines described, MIF (macrophage migration inhibitory factor) was originally shown to inhibit random macrophage migration, but was later found to be chemotactic. With the discovery that MIF is a non-canonical ligand of chemokine receptors, MIF is now proposed to belong to a group of chemokine-like function (CLF) proteins, or "microchemokines". It is an important mediator of the innate immune response with potential roles in the pathophysiology of inflammatory, autoimmune, and neoplastic disorders. The human MIF gene encodes a 115 amino acid, 12.5 kDa secreted protein. Crystallography studies suggest that MIF exists as a homotrimer, but dimers or monomers are also possible. MIF shares minimal homology with other known cytokines. It exhibits intrinsic tautomerase and oxidoreductase activity, but the physiological importance of MIF as an enzyme remains unclear. |
