Background	Aldehyde dehydrogenase 1 family member L1 (ALDH1L1), a 10-formyltetrahydrofolate dehydrogenase that in humans is encoded by the ALDH1L1 gene. The ALDH1L1 protein is a tetramer composed of 902 aa identical subunits. ALDH1L1 catalyzes the conversion of 10-formyltetrahydrofolate, nicotinamide adenine dinucleotide phosphate (NADP+), and water to tetrahydrofolate, NADPH, and carbon dioxide. It belongs to the aldehyde dehydrogenase family. Loss of function or expression of this gene is associated with decreased apoptosis, increased cell motility, and cancer progression. Deficiencies in this gene can result in an accumulation of formate and subsequent methanol poisoning. Many studies have predicted that ALDH1L1 is a tumor suppressor gene. ALDH1L1 is usually underexpressed in malignant tumors and competes with tumors for the same folate substrate. In hepatocellular carcinoma decreased expression of ALDH1L1 is associated with a poor prognosis.
