Background | AMBRA1 (Activating molecule in Beclin1-regulated autophagy protein 1, Kiaa1736, DCAF3) is a regulator of autophagy and neural development in embryogenesis. Ambra1 controls autophagy in the central nervous system by activating class III PtdIns3k complexes that are required for phagophore nucleation. Beclin1 and Vps34 interaction are promoted by AMBRA1, and help form this complex. The loss of AMBRA1 causes upregulated apoptosis and accumulation of ubiquitinated proteins, which finally proves lethal. It was recently discovered that AMBRA1 interacts with Parkin after prolonged mitochondrial depolarization, thus triggering mitophagy (PMID 21921694). AMBRA1 has 7 known isoforms, and numerous WD40 domains. |

Note | This AMBRA1 antibody is useful for Western Blot, Immunocytochemistry/Immunofluorescence, and IHC-paraffin embedded sections. In Western Blot, a band is seen ~ 142 kDa representing AMBRA1. In ICC/IF, cytoplasmic staining was observed in HeLa cells. In IHC-P, staining was observed compartmentalized in cytoplasmic vesicles of mouse brain and skin tissue. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. |
