Recommend Dilution	Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of Rat GROβ/MIP-2 (100 ng/ml), a concentration of 3.0-5.0 µg/ml of this antibody is required ELISA: (Indirect): To detect Rat GROβ/MIP-2 by indirect ELISA (using 100 μl/well antibody solution) a concentration of 0.5-2.0 μg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2- 0.4 ng/well of recombinant Rat GROβ/MIP-2. (Sandwich): To detect Rat GROβ/MIP-2 by sandwich ELISA (using 100 μl/well antibody solution) a concentration of 0.5-2.0 μg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Rat GROβ/MIP-2 (PP032B) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant Rat GROβ/MIP-2. Western Blot: To detect Rat GROβ/MIP-2 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Rat GROβ/MIP-2 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Background	GRO beta is a member of the CXC, or chemokine class. It contains the ELR domain immediately preceding the first cysteine residue near the amino terminus. Other chemokines in this group include IL8, GRO alpha/beta/gamma, mouse KC, ENA78, GCP2, PBP/CTAPIII/beta TG/NAP2. These chemokines act primarily on neutrophils as chemoattractants and activators, including neutrophil degradation with release of myloperoxidase and other enzymes. GRO beta was originally identified as a heparin-binding protein secreted from a murine macrophage cell line in response to endotoxin stimulation. GRO beta is an approximately 8 kDa polypeptide of 73 amino acids. The precursor form of GRO beta consists of 100 amino acids. To generate the mature GRO beta, the precursor cleaves its amino terminal 27 amino acids. GRO beta shows 60% amino acid homology to human GRO alpha and GRO gamma.
