Background E-Cadherin, also called L-CAM (Liver Cell Adhesion Molecule) is an epithelial cell adhesion molecule and plays an important role in the formation of cell-cell contacts in epithelia irrespective of their origin as ecto-, meso- or endodermal tissue. This early adhesion event between epithelial cells is a prerequisite for the assembly of all elements of the junctional complex and is as such important in maintaining epithelial integrity. Over 90% of malignancies are carcinomas. One of the prerequisites for the release of carcinoma cells from the primary site might be a defect in intercellular adhesion mediated by the absence of E-cadherin expression. Therefore, the expression of E-cadherin might be an important parameter for the determination of the invasive potential of epithelial neoplasms, and for the transition of a benign to a malignant neoplasm.

Note Protocol: Protocol with frozen, ice-cold acetone-fixed sections:
The whole procedure is performed at room temperature
1. Wash in PBS
2. Block endogenous peroxidase
3. Wash in PBS
4. Block with 10% normal goat serum in PBS for 30min. in a humid chamber
5. Incubate with primary antibody (dilution see datasheet) for 1h in a humid chamber
6. Wash in PBS
7. Incubate with secondary antibody (peroxidase-conjugated goat anti mouse IgG+IgM (H+L) minimal-cross reaction to human) for 1h in a humid chamber
8. Wash in PBS
9. Incubate with AEC substrate (3-amino-9-ethylcarbazol) for 12min.
10. Wash in PBS
11. Counterstain with Mayer's hemalum
