Preparation and Pretreatment
1. Cut 3-4 μm section of formalin-fixed paraffin-embedded tissue and place on positively charged slides; dry overnight at 58°C.
2. Deparaffinize, rehydrate, and epitope retrieve; the preferred method is the use of Heat Induced Epitope Retrieval (HIER) technique in conjunction with a pressure cooker. The preferred method allows for simultaneous deparaffinization, rehydration, and epitope retrieval. Upon completion, rinse with 5 changes of distilled or deionized water.
3. If using HRP detection system, place slides in peroxide block for 10 minutes; rinse. If using AP detection system, omit this step.

Specificity This antibody detects Smooth Muscle Myosin, heavy chain.
It reacts with Human visceral and vascular smooth muscle cells.
The antibody also reacts with Human myoepithelial cells. It is very helpful in distinguishing between benign sclerosing breast lesions and infiltrating carcinomas in difficult cases since it strongly stains the myoepithelial layer in the benign lesions while it is negative in the infiltrating carcinomas.

Background Smooth Muscle Myosin, heavy chain (SM-MHC, SMMS-1) is a cytoplasmic structural protein that is a major component of the contractile apparatus of the smooth muscle cells. SMMS-1 is also a myoepithelium-associated protein. 
Expression of smooth muscle myosin is developmently regulated, appearing early in smooth muscle development, and is specific for smooth muscle development. Two isoforms of smooth muscle myosin heavy chain have been identified, designated MHC-1 and MHC-2.
