Summary | Herpesvirus entry mediator (HVEM) is a type I transmembrane protein that is a member of the TNF receptor superfamily. It is also known as tumor necrosis factor receptor superfamily member 14 and expressed on various immune cells, including T and B cells, NK cells, monocytes, and neutrophils. Interaction of HVEM with TNF family member LIGHT co-stimulates T cells and promotes inflammation. HVEM also triggers inhibitory signaling cascade in effector T (Teff) cells and regulatory T cells (Tregs) as a ligand of B and T lymphocyte attenuator. Therefore, HVEM is considered as a molecular switch, capable of facilitating both stimulatory and inhibitory cosignaling in T cells. It is found that HVEM Ig can activate a significant antiviral effect against HSV-1 infection. |
