Background | BNIP3L/NIX is a member of the BCL2/adenovirus E1B 19 kd-interacting protein (BNIP) family, which is involved in the regulation of apoptosis. Like other BH3-only proteins, NIX is believed to possess tumor suppressing functions; NIX localizes to the mitochondria and interacts with BCL2 and BCL-X in a pro-apoptotic manner. NIX shares a high level of sequence homology with BNIP3, and like BNIP3 activates apoptosis through its C-terminal transmembrane domain but not via its BH3 domain. Hypoxia is believed to be a principle regulator of NIX activity, making NIX a key factor in hypoxia-induced tumor cell death and heart disease. In addition, NIX has been implicated in mitophagy, a specific form of autophagy; due to its dual role, NIX is an interesting target to examine the relationship between apoptosis and autophagy. |
