Specificity | Recognizes Protein Gene Product 9.5 (PGP9.5). This Monoclonal Antibody reacts with a protein of 20-30kDa, identified as PGP9.5, also known as ubiquitin carboxyl-terminal hydrolase-1 (UchL1). BM160 stains neuronal cell bodies and axons in the CNS and periphery, small nerve fibres in peripheral tissues, neuroendocrine cells in the pituitary, thyroid, pancreas and tumours of the DNES. Also stains neuroendocrine cells in human adult gut (unlike 31A3). Clones 31A3 and 13C4 each recognize a different epitope towards the N-terminus of the protein. Cellular Localization: Cytoplasmic. Background | PGP9.5 is a ubiquitin hydrolase which is widely expressed in neuronal tissues and represents 1-2% of total soluble brain proteins. PGP9.5, also known as ubiquitin C-terminal hydrolase 1 (UCHL-1), is involved in the regulation of the ubuiquitin pathway. Initially, PGP9.5 expression in normal tissues was reported in neurons and neuroendocrine cells but later it was found in distal renal tubular epithelium, spermatogonia, Leydig cells, oocytes, melanocytes, prostatic secretory epithelium, ejaculatory duct cells, epididymis, mammary epithelial cells, Merkel cells, and dermal fibroblasts. Furthermore, immunostaining for PGP9.5 has been shown in a wide variety of mesenchymal neoplasms as well. A mutation in PGP9.5 gene is believed to cause a form of Parkinson's disease. |
