Reconstitution | 1. Centrifuge at 5,000xg for 5min. 2. Carefully open the tube and add 100ul of sterile water to dissolve the DNA. 3. Close the tube and incubate for 10 minutes at room temperature. 4. Briefly vortex the tube and then do a quick spin (less than 5000xg) to concentrate the liquid at the bottom. 5. Store the suspended plasmid at -20°C. The DNA is stable for at least one year from date of shipping when stored at -20°C. |

Gene Summary | This gene encodes a serine/threonine-protein kinase that is involved in mitotic regulation. This protein is localized to the centrosome, and undetectable during G1 phase, but accumulates progressively throughout the S phase, reaching maximal levels in late G2 phase. Alternatively spliced transcript variants encoding different isoforms with distinct C-termini have been noted for this gene. [provided by RefSeq, Feb 2011] Transcript Variant: This variant (1) represents the predominant transcript, and encodes the longest isoform (1, also known as NEK2A). The C-terminus (aa 399-445) of this isoform was shown to be responsible for its nucleolar localization (PMID:15161910). Isoforms 1 and 2 (which differ at the C-terminus) have also been reported to exhibit distinct pattern of expression during mitosis (PMID:11742531). |
